Structure of Macronuclear Envelopes of Tetrahymena Pyriformis in the Stationary Phase of Growth

چکیده
In a preceding publication a method was reported by which fragments of the nuclear envelopes of Tetrahymena pyriformis could be isolated (10). When macronuclear envelopes were isolated from exponentially growing cultures of the amicronucleate Tetrahymena pyriformis strain GL, two types of envelope pieces were observed which differed markedly in the structure of their pore complexes. Type A contained pores with a diameter in the range of 55-60 mp and showed almost no annulus material; whereas, the pores of type B possessed an extremely broad annulus with an inner annulus diameter of about 20 mgl. The nuclear pore complex, according to a general hypothesis, is thought of as a site of processes which regulate nucleocytoplasmic interaction (47, 21), e.g. those controlling the passage of macromolecules involved in cellular protein synthesis (11, 22, 26). Therefore, it is of particular interest to compare the structure of the macronuclear pores from exponentially growing cells with that of the macronuclear pores from cells in the stationary phase, i.e. under conditions in which the nuclear activity is minimal (27). Whereas the previous study (10) reported the structure of macronuclear membranes from log phase Tetrahymena cells, the present study deals with the pore complex of stationary phase cells.

The Fate of Mitochondria during Aging in Tetrahymena Pyriformis
During the growth cycle of Tetrahymena pyriformis the mitochondria undergo changes in position, number, and structure. Ciliates in the logarithmic growth phase possess elongated mitochondria which are aligned along the plasma membrane and are closely associated with the kinetosomes and kinetodesmata. Mitochondria appear to divide across the long axis at this time, resulting in two or more produ...

Histone composition of a chromatin fraction containing ribosomal deoxyribonucleic acid isolated from the macronucleus of Tetrahymena pyriformis.
The histone compositions of a chromatin fraction containing ribosomal DNA and of the remaining macronuclear chromatin of Tetrahymena pyriformis was analysed by gel electrophoresis. These chromatin fractions were used as models for transcriptionally active and inactive chromatin respectively. The extent of histone modification, as indicated by the distribution of histone between differently char...
متن کاملStudies on Nuclear Structure and Function in Tetrahymena Pyriformis
This report describes a rapid, efficient method for isolating macronuclei from Tetrahymena. The macronuclear fraction contains only small amounts of micronuclear material and little detectable whole cell or cytoplasmic contamination. A method is also described for preparing a "micronuclear fraction" which contains 20-40 micronuclei for every macronucleus present. Electron microscope observation...
متن کامل[6n]methyl Adenine in the Nuclear Dna of a Eucaryote, Tetrahymena Pyriformis
DNA isolated from macronuclei of the ciliate, Tetrahymena pyriformis, has been found to contain [(6)N]methyl adenine (MeAde); this represents the first clear demonstration of significant amounts of MeAde in the DNA of a eucaryote. The amounts of macronuclear MeAde differed slightly between different strains of Tetrahymena, with approximately 0.65-0.80% of the adenine bases being methylated. The...
